چکیده

A rapid and sensitive assay for quantification of nalbuphine, butorphanol and morphine in blood (50μl) and brain microdialysate (~40μl) samples was developed. Blood samples were extracted with ethyl acetate. Analysis was performed with high performance liquid chromatography coupled to an electrochemical detector. The mobile phase was a mixture of 0.1M sodium phosphate buffer, methanol and octane-sulfonic acid with ratio and pH depending on compound and matrix. The limits of quantification in blood samples were 25, 50 and 25 ng/ml for nalbuphine, butorphanol and morphine, respectively and 0.5 ng/ml for morphine in microdialysate samples. Based on sample volume, sensitivity and reproducibility, these assays are particularly suitable for pharmacokinetic-pharmacodynamic studies in rodents.

Modelling of the Eeg Effects of Opioids

The objective of this investigation was to characterize the role of complex biophase distribution kinetics in the pharmacokinetic-pharmacodynamic correlation of a wide range of opioids. Following intravenous infusion of morphine, alfentanil, fentanyl, sufentanil, butorphanol and nalbuphine the time course of the EEG effect was determined in conjunction with blood concentrations. Different bioph...

Mechanism-based pharmacokinetic-pharmacodynamic modeling of the respiratory-depressant effect of buprenorphine and fentanyl in rats.

The purpose of this investigation was to develop a mechanism-based pharmacokinetic/pharmacodynamic (PK/PD) model to predict the time course of respiratory depression following administration of opioids in rats. The proposed model is based on receptor theory and aims at the separate characterization of biophase distribution and receptor association/dissociation kinetics as determinants of hyster...

Conformational study of catecholamines in solution.

Conformations of noradrenaline, dopamine and ephedrine have been studied in aqueous solutions using high resolution PMR spectroscopy. We found that for dopamine the mole fraction of trans rotamer is increased if pH is changed from acidic to basic while for noradrenaline and ephedrine the opposite trend is observed: gauche rotamer forms of the side chain become more populated. These changes are ...

Investigating the Effect of Topographic factors on the distribution of oriental beech in Hyrcanian forest, Iran

Studying the present and future distribution status of important forest species is a prerequisite for the need of conservation programs in order to play their undeniable role in tackling global climate change, land use change and air pollution. The purpose of this study was to determine the distribution of oriental beech (Fagus orientalis Lipsky) in forests of northern Iran under the influence ...

O-44: Characterisation of Monotreme CaseinsReveals Lineage Specific Expansion of an AncestralCasein Locus in Mammals

Background: One important reproductive characteristic of Mammals is the production of milk to nurse the neonate. In order to better understand the evolution of milk we have investigated gene expression in milk cells from monotremes which are the most ancient representative of the mammalian lineage. Materials and Methods: Using a milk cell cDNA sequencing approach we characterise milk protein se...
